文档编号:SW032 文档字数:11089,页数:31 摘要:本文档研究桑葚中生物类总黄酮的提取条件与大孔吸附树脂分离与纯化总黄酮的工艺条件。结合正交实验设计方法,以桑葚中生物类总黄酮的提取率为考察指标,对影响桑葚中生物类总黄酮提取的乙醇质量分数,提取时间,提取温度,料液比进行研究;以桑葚中总黄酮吸附量、解吸量为指标,选择合适的大孔吸附树脂,并研究吸附与解吸优化条件,对影响分离纯化的pH值,无机盐浓度,温度,乙醇浓度等进行了分析。桑葚中生物类总黄酮提取的最优条件,即:以75%乙醇为提取剂,提取温度75℃,料液比为1:15,提取时间为2 h,再结合分离纯化的较优条件,条件为:待分离样品溶液pH值调至3.0-4.0左右,无机盐浓度调至3%-6%,吸附的温度为50℃,洗脱最佳溶剂为50%的乙醇溶液,洗脱速率为0.5-1.0ml/min,洗脱用量为3倍的柱体积。桑葚中生物类总黄酮的提取率可以达到 2600mg/kg(以芦丁计),桑葚提取物总黄酮的含量接近50%。此正交实验设计可行,选取D-101型树脂为合适大孔吸附树脂,选取药桑葚或黑桑葚为佳,D-101型大孔吸附树脂适用于桑葚中总黄酮的分离与纯化。 关键词:桑葚,总黄酮,正交设计,大孔吸附树脂,分离纯化 Abstract: To study technique condition for separating total flavonoids from mulberry. With orthogonal experiment based on single factor, The total extraction rate of flavonoids for purpose, the effects of ethanol concentration, ratio of material to liquid, extraction temperature, extraction temperature, extraction time and times on extraction of total flavonoids from mulberry were study to obtain the optimal extraction conditions; adsorbance of mulberry total flavonoids for indicators, Choose a suitable macroporous resin, Study Adsorption and Desorption optimum conditions, analysis pH, salt concentration, temperature, ethanol concentration. Mulberry of total flavonoids extracted from optimal conditions, 75% of ethanol, water bath temperature of 75 ℃, extracting liquid ratio of 1:15, extraction time is 2h, pH is 4.0 , salt oconcentration is 3% , temperature is 50℃, ethanol concentration is 50%, elution rate of ethanol is 0.5-1.0ml/min,3 column volume. Mulberry of total flavonoids in the extraction rate can be achieved 2600mg/kg. Under the experimental conditions, total flavonoids extracted from mulberry by D-101 macroporous resin nearly 50%. Choose the D-101macroporous resin, choose the medicine mulberry or black mulberry, and D-101 macroporous resin applied to mulberry of total flavonoids in the separation and purification.
