文档编号:SW071 文档字数:9939,页数:26 摘 要:近年来,随着绿色化学的兴起和对节能、环保的重视,在非水相反应体系中使用固定化酶不但能在一定程度上减轻溶剂对酶的失活作用,而且固定化酶的分散更为均匀,避免了酶的聚集,易于处理。本实验以毛巾为载体,用戊二醛进行吸附交联固定脂肪酶,并利用固定化脂肪酶在非水相中催化正丁酸和乙醇的合成反应。研究了固定化过程中载体质量、酶加载量,反应体系中的时间、底物浓度及摩尔比、含水量、表面活性剂、储存和温度稳定性等因素对催化丁酸乙酯反应的影响规律,确定最优条件:在0.6g载体上固定脂肪酶0.1g/10ml为宜,在反应体系中乙醇和丁酸最适比例约为1:2,游离酶最适含水量为O.5%。表面活性剂中Tween60对酶活性的提升效果最为明显,另外,在时间、稳定性研究中对底物和产物的趋势亦作了讨论。 关键词:脂肪酶;固定化;酯催化;丁酸乙酯 Catalytic synthesis of ethyl butyrate by immobilized lipase Abstract: In recent years, with the rise of green chemistry and energy-saving, environmental protection, enzyme reuse has become an important topic of the biological engineering, enzyme immobilization is one of the methods to solve this problem, In no water system, immobilized enzyme system not only can alleviate deactivation function of solvent for enzyme, but also can scatter more evenly in the system, which makes it easy to handle.
